Our tissue processing and single cell library construction protocols are compatible with most human and murine tissue types. We have experimentally verified more than 130 sample types, including both tumors and healthy tissues of lung, liver, stomach, thyroid, brain, ovary, endometrial tissue, testis, prostate, bone, and blood. A successful tissue dissociation procedure should typically generate single cell suspension with more than 85% of living cells and free of aggregates and debris.
We do not recommend such experiments. Low cell viability normally indicates adverse conditions that could have affected all cells in the same sample. The effects could include RNA degradation and/or change of gene expression patterns, making single cell RNA-seq results less reliable. If the sample is rare and difficult to get, we can offer additional procedure to remove dead cells before proceed to single cell library construction and sequencing.
