Single cell RNA sequencing normally requires single cell suspensions from fresh samples. This can be challenging for certain sample types such as archived samples that are stored frozen tissue. Single nucleus RNA sequencing technology has been developed to sequence mRNA present in single cell nuclei. Nuclei from frozen tissues can be extracted and processed in the single cell sequencing workflow, making it feasible to analyze archived frozen tissue sample by single cell sequencing. This can be especially useful for analyzing clinical samples.
Single Nucleus RNA Sequencing Example
Single nuclei were extracted from frozen mouse kidney and sequenced. Major cell types were successfully identified based on expression profiles of marker genes, as shown in the figure and table above.
